阿托伐他汀對(duì)糖尿病大鼠胸主動(dòng)脈TLR4表達(dá)影響的實(shí)驗(yàn)研究

目的觀察GK大鼠胸主動(dòng)脈TLR4表達(dá)及應(yīng)用阿托伐他汀其表達(dá)水平的變化。探討TLR4與動(dòng)脈硬化的關(guān)系。方法分組:空白對(duì)照組;實(shí)驗(yàn)對(duì)照組;阿托伐他汀組,5周后應(yīng)用RT-PCR、Western blot檢測(cè)各組大鼠胸主動(dòng)脈TLR4表達(dá)。結(jié)果實(shí)驗(yàn)對(duì)照組較空白對(duì)照組TLR4表達(dá)顯著增高,阿托伐他汀組較實(shí)驗(yàn)對(duì)照組TLR4表達(dá)顯著降低。結(jié)論阿托伐他汀可能通過(guò)降低TLR4表達(dá),防治動(dòng)脈粥樣硬化。

Toll樣受體是一組跨膜受體蛋白質(zhì)[1],TLR4與AS的關(guān)系受到廣泛關(guān)注。本研究通過(guò)阿托伐他汀干預(yù)GK大鼠,觀察TLR4表達(dá)的變化,探討TLR4在糖尿病血管粥樣硬化發(fā)生、發(fā)展中的作用,為動(dòng)脈粥樣硬化防治提供新的切入點(diǎn)。
1.1 實(shí)驗(yàn)動(dòng)物:GK大鼠,Wister大鼠。
1.2 實(shí)驗(yàn)方法:分組:①空白對(duì)照組:Wister大鼠8只;②實(shí)驗(yàn)對(duì)照組:GK大鼠8只,蒸餾水灌胃;③阿托伐他汀組:GK大鼠8只,阿托伐他汀50 mg(kg?d)灌胃。5周后3%戊巴比妥鈉麻醉大鼠,取胸主動(dòng)脈組織,制備組織研漿。RT-PCR及Western blot檢測(cè)TLR4表達(dá)。
1.3 統(tǒng)計(jì)學(xué)方法:SPSS13.0軟件分析數(shù)據(jù),采用t檢驗(yàn),P<0.01有統(tǒng)計(jì)學(xué)意義。

動(dòng)脈粥樣硬化是一個(gè)炎癥過(guò)程[2]。糖尿病是動(dòng)脈粥樣硬化的獨(dú)立危險(xiǎn)因素[3]。糖尿病與動(dòng)脈粥樣硬化均為炎癥性疾病[4],TLR4為介導(dǎo)炎性反應(yīng)介子,抑制TLR4表達(dá)可能為抗動(dòng)脈硬化的機(jī)制之一,本研究發(fā)現(xiàn)實(shí)驗(yàn)對(duì)照組大鼠胸主動(dòng)脈TLR4表達(dá)顯著高于空白對(duì)照組,阿托伐他汀治療后可以抑制TLR4表達(dá),具有抑制動(dòng)脈粥樣硬化的作用。通過(guò)研究,我們認(rèn)為TLR4可能在糖尿病大鼠動(dòng)脈硬化的發(fā)生發(fā)展中扮演重要角色,阿托伐他汀具有抗炎作用,可能通過(guò)不同的機(jī)制抑制TLR4的表達(dá),而具有防治糖尿病性動(dòng)脈硬化作用,從而改善糖尿病血管病變。

Experimental Study of the Effect of Atorvastatin on the Expression of TLR4 in Aorta Thoracalis of the GK Mice

ObjectiveTo observe the expression of TLR4 in aorta thoracalis of the GK mice and the change of that after using atorvastatin.MethodsMice were divided into control group, experimental control group, atorvastatin group. After fi ve weeks, TLR4 was detected by using PT-PCR and Western blot method.ResultsThe TLR4 level of experimental control group is higher than that of control group. The TLR4 level of atorvastatin group is lower than that of experimental control group(P<0.01).ConclusionsAtorvastatin may prevent atherosclerosis through reducing TLR4 expression.
