警惕新德里番茄曲葉病毒在我國的傳播和危害

摘要

新德里番茄曲葉病毒(tomato?leaf?curl?New?Delhi?virus,?ToLCNDV)是雙生病毒科菜豆金色花葉病毒屬病毒,由煙粉虱Bemisia?tabaci以循環(huán)持久性方式傳播,能對茄科和葫蘆科等多種作物造成毀滅性的危害。近年來,ToLCNDV不斷擴(kuò)散蔓延至新的國家和地區(qū)。我國于2021年在浙江的溫室番茄上發(fā)現(xiàn)了ToLCNDV的危害。本文綜述了ToLCNDV的發(fā)生分布、基因組結(jié)構(gòu)與進(jìn)化、傳播方式和寄主范圍,提出了防范ToLCNDV在我國進(jìn)一步傳播和危害的防控建議。

新德里番茄曲葉病毒(tomato?leaf?curl?New?Delhi?virus,?ToLCNDV)是一種對番茄Solanum?lycopersicum等作物造成毀滅性危害的雙生病毒。該病毒最早于1995年在印度發(fā)現(xiàn)[1],而后迅速蔓延至印度的大多數(shù)地區(qū)以及巴基斯坦、孟加拉國、泰國等亞洲國家。2012年以來,ToLCNDV向西傳播擴(kuò)散至伊朗和西班牙等國,對黃瓜Cucumis?sativus、西葫蘆Cucurbita?pepo和西瓜Citrullus?lanatus等葫蘆科作物造成嚴(yán)重危害,被歐洲和地中海植物保護(hù)組織(European?and?Mediterranean?Plant?Protection?Organization,EPPO)列入警戒名單[24]。2021年Li等在我國浙江的溫室番茄上發(fā)現(xiàn)了ToLCNDV的危害[5]。鑒于ToLCNDV的媒介昆蟲煙粉虱Bemisia?tabaci?在我國廣泛發(fā)生[6],需高度警惕該病毒在我國的發(fā)生與蔓延。本文綜述了ToLCNDV的發(fā)生分布、基因組結(jié)構(gòu)與進(jìn)化關(guān)系、傳播方式和危害特征等內(nèi)容,并提出了防范ToLCNDV在我國進(jìn)一步傳播和危害的建議。

1?ToLCNDV的發(fā)生分布

ToLCNDV最早于1995年在印度被報(bào)道危害番茄[1]。1996年-1998年,研究者們發(fā)現(xiàn)除了番茄外,ToLCNDV還危害泰國的甜瓜Cucumis?melo、黃瓜、葫蘆Lagenaria?siceraria和絲瓜Luffa?aegyptiaca以及巴基斯坦的龍葵Solanum?nigrum[4,7]。與此同時(shí),ToLCNDV擴(kuò)散到印度各個(gè)地區(qū),寄主范圍不斷擴(kuò)大[89]。隨后,該病毒被報(bào)道在印度和巴基斯坦危害胡蘿卜Daucus?carota?var.?sativa和苦瓜Momordica?charantia等作物[3,1011]。2012年-2013年,ToLCNDV被證實(shí)與西班牙葫蘆曲葉病相關(guān)[12]。2013年-2015年,巴基斯坦報(bào)道了ToLCNDV與棉花曲葉病的發(fā)生相關(guān)[13]。隨后,ToLCNDV被報(bào)道在突尼斯的黃瓜、甜瓜和西葫蘆以及意大利的西葫蘆上引起了嚴(yán)重的危害[1415]。2021年,我國浙江約0.5?hm2的溫室番茄出現(xiàn)葉片卷曲的癥狀,經(jīng)高通量測序和分子生物學(xué)鑒定,發(fā)現(xiàn)引起該病害的病原是ToLCNDV[5]。

根據(jù)EPPO的最新數(shù)據(jù),ToLCNDV已經(jīng)在亞洲、非洲、歐洲和地中海地區(qū)的19個(gè)國家被發(fā)現(xiàn)并報(bào)道[13,5,7,12,1424]?(表1)。

2?ToLCNDV的基因組結(jié)構(gòu)與系統(tǒng)進(jìn)化關(guān)系

ToLCNDV是一種雙組分的菜豆金色花葉病毒屬病毒,基因組為單鏈環(huán)狀DNA,包含兩個(gè)大小均約為2.7?kb的DNAA和DNAB組分(圖1)。DNAA組分含有6個(gè)ORF,分別為病毒鏈的AV1和AV2以及互補(bǔ)鏈的AC1、AC2、AC3和AC4。AV1編碼病毒的外殼蛋白,AV2可能作為RNA沉默抑制子抑制RNA沉默介導(dǎo)的防御反應(yīng),AC1編碼病毒復(fù)制相關(guān)蛋白,AC2編碼轉(zhuǎn)錄激活蛋白,AC3編碼復(fù)制增強(qiáng)蛋白,AC4是癥狀決定因子和RNA沉默抑制子。DNAB組分含有2個(gè)ORF,即病毒鏈的BV1和互補(bǔ)鏈的BC1,分別編碼病毒的核穿梭蛋白和運(yùn)動(dòng)蛋白。DNAA和DNAB的病毒鏈和互補(bǔ)鏈由共同區(qū)(common?region,?CR)隔開,CR包含病毒復(fù)制所需的莖環(huán)結(jié)構(gòu)和9堿基保守序列(TAATATT↓AC)。與大多數(shù)的雙生病毒相似,ToLCNDV編碼的大多數(shù)蛋白具有多功能的特性,通過利用寄主因子完成復(fù)制、轉(zhuǎn)錄、運(yùn)動(dòng)等生活史或通過不同方式逃脫植物的抗病毒防御反應(yīng)而促進(jìn)病毒侵染[2528]。

對ToLCNDV?DNAA的全長序列進(jìn)行分析,發(fā)現(xiàn)ToLCNDV的序列具有較高的遺傳多樣性。根據(jù)國際病毒分類委員會(huì)的分類建議[29],現(xiàn)有的ToLCNDV至少可以分為7個(gè)不同的株系。大多數(shù)來源于亞洲的ToLCNDV分離物聚為一類,相似性在94%以上;來源于地中海地區(qū)的ToLCNDV分離物聚為一類,序列相似性在99%以上,但是與目前報(bào)道的其他ToLCNDV分離物相比,序列相似性在94%以下,被認(rèn)為是ToLCNDV新株系,命名為ToLCNDVES。此外,一部分來源于印度和巴基斯坦的ToLCNDV分離物聚為另外一類[34]。重組是驅(qū)動(dòng)雙生病毒進(jìn)化的重要因素,分析發(fā)現(xiàn)ToLCNDV的不同株系是由ToLCNDV與別的病毒重組產(chǎn)生的[3]。

3?ToLCNDV的傳播方式

ToLCNDV在自然條件下由煙粉虱以循環(huán)持久性方式傳播,即煙粉虱取食帶毒植株后病毒隨唾液進(jìn)入到腸道,滲透過腸壁進(jìn)入血淋巴,最后循環(huán)回到唾液腺,再取食其他植物時(shí)病毒隨唾液進(jìn)入植物體內(nèi)。雖然ToLCNDV不能在煙粉虱體內(nèi)增殖,但是煙粉虱一旦獲毒可終身帶毒并傳毒。病毒除隨煙粉虱傳播,也可通過寄主和非寄主植物以及切花和新鮮草藥等材料,經(jīng)國際貿(mào)易遠(yuǎn)距離傳播。與其他雙生病毒相比,ToLCNDV與煙粉虱互作方面的研究報(bào)道還比較少。另有報(bào)道表明,感染ToLCNDV的西葫蘆可以通過帶毒種子傳播[3031],但也有研究發(fā)現(xiàn)感染ToLCNDV的甜瓜種子可帶毒,但是不傳播病毒[32]。

4?ToLCNDV的危害特征

ToLCNDV寄主范圍很廣,可侵染茄科Solanaceae、葫蘆科Cucurbitaceae、番木瓜科Caricaceae、豆科Fabaceae、錦葵科Malvaceae和罌粟科Papaveraceae等17科55種作物、花卉和雜草[2],常見的寄主有番茄、西葫蘆、黃瓜、西瓜等茄科和葫蘆科植物?(表2)。ToLCNDV危害番茄主要引起葉片卷曲、皺縮、脈明和黃斑駁等癥狀,在侵染后期引起節(jié)間縮短,影響坐果和產(chǎn)量(圖2a);危害南瓜等葫蘆科作物引起葉片卷曲、矮縮等癥狀(圖2b)。ToLCNDV的不同分離物對不同寄主植物的適應(yīng)性以及所導(dǎo)致的癥狀可能不同[34]。例如,ToLCNDV印度分離物在番茄上具有很強(qiáng)的適應(yīng)性并誘導(dǎo)番茄植株產(chǎn)生明顯的癥狀,但是來源于地中海地區(qū)的ToLCNDVES株系主要危害葫蘆科作物,而對番茄的侵染性很低[12,33]。在自然界中,ToLCNDV可與雙生病毒衛(wèi)星DNA如alpha衛(wèi)星和beta衛(wèi)星相結(jié)合。Alpha衛(wèi)星和beta衛(wèi)星大小均約1.3?kb。Beta衛(wèi)星是很多單組分雙生病毒誘導(dǎo)寄主產(chǎn)生癥狀所必需的,其互補(bǔ)鏈上編碼的βC1蛋白可通過不同作用方式從DNA水平、RNA水平到蛋白質(zhì)水平破壞寄主的抗病毒防御反應(yīng)而促進(jìn)病毒侵染[34]。模擬接種試驗(yàn)發(fā)現(xiàn),beta衛(wèi)星加重ToLCNDV侵染后寄主的癥狀并且擴(kuò)大ToLCNDV的寄主范圍[3537]。ToLCNDV還與其他雙生病毒如番茄黃曲葉病毒(tomato?yellow?leaf?curl?virus,?TYLCV)、澤蘭黃脈花葉病毒(bhendi?yellow?vein?mosaic?virus)、棉花曲葉病毒(cotton?leaf?curl?virus)等病毒復(fù)合侵染,引起更加嚴(yán)重的癥狀或者產(chǎn)生新的致病類型[13,?34,?3841]。

5?監(jiān)測與防控

ToLCNDV在印度發(fā)現(xiàn)并對印度番茄具有極大的破壞性。近年來該病毒已經(jīng)傳播擴(kuò)散至不同的國家和地區(qū),對茄科和葫蘆科等作物的生產(chǎn)造成了嚴(yán)重影響。與ToLCNDV相似的番茄黃曲葉病毒(TYLCV)也屬于雙生病毒科菜豆金色花葉病毒屬,可由煙粉虱傳播。TYLCV自2006年首次入侵我國以來,已在我國24個(gè)省、直轄市、自治區(qū)危害番茄、辣椒和煙草等25種植物,造成巨大損失[42]。鑒于該類病毒的傳播特性和煙粉虱在我國的廣泛分布,且發(fā)生后基本無藥劑可用的現(xiàn)狀,需遵循“預(yù)防為主、綜合防控”的植保方針[43],強(qiáng)化對ToLCNDV的檢疫監(jiān)測預(yù)警,明確ToLCNDV的侵染循環(huán)和流行規(guī)律,加強(qiáng)抗性品種選育研究,控制ToLCNDV在我國的傳播和危害。

5.1?加強(qiáng)對ToLCNDV的檢疫預(yù)警

鑒于該病毒威脅巨大,土耳其等國已通過WTO/SPS評議對進(jìn)口的寄主植物種子提出了檢疫要求(http:∥www.tbtsps.gov.cn/tbcx/getTbcxContent.action?mid=27178&TBType=5),預(yù)計(jì)還將有更多的國家將其列入檢疫性有害生物管理。我國可根據(jù)目前ToLCNDV的發(fā)生和分布信息,開展風(fēng)險(xiǎn)分析,明確ToLCNDV在我國的適生區(qū)和潛在危害損失,評估將其作為我國進(jìn)境和全國農(nóng)業(yè)植物檢疫性有害生物管理的必要性和可行性。

5.2?加強(qiáng)對ToLCNDV的監(jiān)測與防控

開展普查和重點(diǎn)監(jiān)測,明確ToLCNDV在我國的發(fā)生分布范圍。強(qiáng)化對重點(diǎn)種子苗木基地的監(jiān)測,開發(fā)快速檢測試劑盒或者試紙條,在病害發(fā)生初期盡早和精準(zhǔn)檢測出病毒,提升末端早發(fā)現(xiàn)、早處置的能力。此外,輪作以及避開煙粉虱發(fā)生高峰期種植作物對于控制ToLCNDV的發(fā)生有一定效果。在沒有煙粉虱或者在保護(hù)性設(shè)施中煙粉虱種群數(shù)量低的情況下,拔除感染病毒的植株。在發(fā)生區(qū),可使用防蟲網(wǎng)、黃色粘蟲板等物理屏障驅(qū)避煙粉虱,并且結(jié)合生物防治和化學(xué)防治等有害生物綜合治理策略來降低煙粉虱的種群數(shù)量,從而抑制病毒的傳播[44]。

5.3?選育抗性品種

從長遠(yuǎn)來說,ToLCNDV抗性品種的培育是一項(xiàng)艱巨的工作,但卻是控制ToLCNDV最經(jīng)濟(jì)有效的手段。近期研究發(fā)現(xiàn),抗性基因Ty2和Ty3聯(lián)合使用能夠增強(qiáng)番茄對ToLCNDV的抗性[45],Sw5a能夠提高番茄對ToLCNDV的抗性[4647]。下一步,需要對我國廣泛種植的茄科和葫蘆科作物的抗性進(jìn)行評估,加強(qiáng)對ToLCNDV的致病機(jī)制和植物抗病毒機(jī)制的研究,挖掘針對ToLCNDV的抗性基因,為利用基因工程技術(shù)研發(fā)培育ToLCNDV的抗性品種提供靶標(biāo)和策略。
