貴州麻陽(yáng)河自然保護(hù)區(qū)7種野生紅菇的形態(tài)與分子鑒定

貴州麻陽(yáng)河自然保護(hù)區(qū)7種野生紅菇的形態(tài)與分子鑒定

為了解麻陽(yáng)河自然保護(hù)區(qū)野生紅菇真菌資源狀況,從保護(hù)區(qū)采集到7種野生紅菇,基于其形態(tài)特征結(jié)合ITS rDNA序列與GenBank中相關(guān)的紅菇屬的同類序列構(gòu)建的分子系統(tǒng)發(fā)育樹,將其鑒定為厭味紅菇Russulanauseosa、美紅菇Russulapuellaris、鱗蓋紅菇Russulalepida、菱紅菇Russulavesca、以及變綠紅菇Russulavirescens、Russulafaustiana、粉柄紅菇Russulafarinipes。

紅菇屬RussulaPers.(1796)在分類學(xué)上隸屬于擔(dān)子菌門Basidiomycota、擔(dān)子菌綱Basidiomycetes、紅菇目Russulales Kreisel ex P.M.Kirk ,P.F.Cannon & J.C.David、紅菇科Russulaceae Lotsy,在真菌系統(tǒng)學(xué)研究中具有相當(dāng)重要意義,也是系統(tǒng)發(fā)育中頗為重要的類群[1-2]。目前國(guó)際上已經(jīng)發(fā)表的紅菇名稱有2600多個(gè)(www.indexfungorum.org,2016),約750多種,但因?yàn)榧t菇屬真菌形態(tài)差異大,導(dǎo)致有許多同種異名或異名同種的現(xiàn)象發(fā)生,修改訂正后,報(bào)道的紅菇屬真菌達(dá)300多種,而我國(guó)記載的就有90余種,其中可食用的有60多種[3]。宋斌等[4]通過(guò)收集并整理文獻(xiàn),匯總了中國(guó)的紅菇屬RussulaPers.真菌,包括179個(gè)名稱記錄。此外,調(diào)查表明,紅菇屬真菌廣泛分布于我國(guó)的30個(gè)省區(qū),在已報(bào)道的紅菇屬名稱記錄中,有效名稱有159個(gè),其中包括148個(gè)種、9個(gè)變種和2個(gè)變型。張?chǎng)蔚萚5]以顯微生態(tài)學(xué)為基礎(chǔ),報(bào)道了亞洲新紀(jì)錄種蠟味紅菇;付立忠等[6]利用分子系統(tǒng)學(xué)鑒定方法,對(duì)采自浙江的四個(gè)紅菇科菌株進(jìn)行測(cè)序分析,以鑒定其親緣關(guān)系;Das等[7]基于ITS基因序列構(gòu)建的系統(tǒng)發(fā)育樹,結(jié)合宏觀和微觀形態(tài)鑒定,報(bào)道了RussulashingbaensisK. Das & S.L. Mill.和RussulaThindiiK. Das & S.L. Mill.,它們是兩個(gè)采自印度的紅菇屬新種。本研究通過(guò)形態(tài)學(xué)和ITS rDNA序列相結(jié)合的方法,對(duì)采自貴州省麻陽(yáng)河自然保護(hù)區(qū)的七種紅菇真菌進(jìn)行了分類鑒定。

1 材料與方法

1.1 材料

本次試驗(yàn)的大型真菌實(shí)體標(biāo)本,均于2013年10月2~5日期間在麻陽(yáng)河自然保護(hù)區(qū)采集,并保存于貴州大學(xué)貴州省生化工程中心標(biāo)本室。

1.2 試劑及儀器

真菌基因組提取試劑盒(Fungal gDNA Kit)(BIOMIGA公司),PCR擴(kuò)增所用試劑[DNA Marker(廣州東盛生物科技有限公司),50×TAE溶液(Solarbio),2×PCR Master Mix、ITS 1、ITS 4、ddH2O(上海前塵生物科技有限公司)。

數(shù)碼相機(jī)(Nikon),顯微鏡(OLYMPUS BX41TF),凝膠成像系統(tǒng)以及PCR儀(Bio-Rad公司)等。

1.3 紅菇的形態(tài)鑒定

根據(jù)記錄本的記錄及照片對(duì)標(biāo)本的宏觀形態(tài)進(jìn)行整理、描述,描述的內(nèi)容包括研究標(biāo)本的生境、習(xí)性;菌蓋的大小、形狀、顏色;菌柄的長(zhǎng)、直徑、形狀等;菌褶生長(zhǎng)情況等。顏色的記錄主要根據(jù)野外記錄情況及拍攝的彩色照片,需參考相關(guān)的工具書,并結(jié)合國(guó)際色彩標(biāo)準(zhǔn)名稱與色值的電子版描述子實(shí)體顏色[8-9]。

1.4 紅菇的分子鑒定

1.4.1 DNA提取

將紅菇研磨成粉末狀,取約10~50 mg,采用真菌基因組提取試劑盒提取紅菇子實(shí)體的DNA,方法按照說(shuō)明書進(jìn)行。

1.4.2 PCR擴(kuò)增和測(cè)序

以適量基因組DNA為模板,進(jìn)行PCR擴(kuò)增,擴(kuò)增引物為ITS 1:5’-3’(TCC GTA GGT GAA CCT GCG G)和ITS 4:5’-3’(TCC TCC GCT TAT TGA TAT GC)[10]。PCR擴(kuò)增的反應(yīng)體系采用25 μL體系,包括有10×擴(kuò)增緩沖液(含MgCl2)2.5μL,200μm dNTP 2.5 μL,5 μm的引物各1 μL,Taq DNA 聚合酶0.3 μL(2.5 u/μL),DNA 模板1.0~2 μL,用ddH2O補(bǔ)至25μL。PCR 擴(kuò)增反應(yīng)在PCR儀上進(jìn)行。擴(kuò)增程序具體參數(shù)如下:95℃預(yù)變性4 min,然后:94℃變性30 s,50℃(有時(shí)55℃)退火40 s,72℃延伸90 s,持續(xù)35個(gè)循環(huán),最后再72℃延伸10 min,于4℃保存。經(jīng)凝膠電泳檢測(cè)后,適合測(cè)序的PCR產(chǎn)物送上海立菲生物技術(shù)有限公司測(cè)序。

1.4.2 序列比對(duì)及分子系統(tǒng)學(xué)分析

使用NCBI中Blast在線檢索,下載與研究樣品相近的ITS rDNA參考序列菌株信息(見表1),采用MEGA5.05[11]分別比對(duì)(alignment)測(cè)得的各個(gè)樣本的基因片段與在GenBank比對(duì)后下載的相關(guān)真菌序列。再用MEGA5.05手工校正來(lái)實(shí)現(xiàn)排序匹配的最優(yōu)化。比對(duì)后的序列采用PAUP* 4.0軟件以最大簡(jiǎn)約法(Maximum parsimony, MP)進(jìn)行分析,以啟發(fā)式搜索法(Heuristic Search)獲得系統(tǒng)發(fā)育樹。其中,啟發(fā)式搜索采用二等分再連接法(Tree-Bisection-Reconnection, TBR)作為獲取聚類樹的方法。在系統(tǒng)發(fā)育分析中堿基序列的空缺(Gaps)作為堿基缺失處理;把所有的堿基狀態(tài)處理為無(wú)序,并給以相同的權(quán)重(轉(zhuǎn)換和顛換的比率為1∶1),即不加權(quán)(Equal weight)處理。經(jīng)1000次重復(fù)計(jì)算自展檢驗(yàn)的值(bootstrap value)以評(píng)價(jià)系統(tǒng)發(fā)育樹拓?fù)浣Y(jié)構(gòu)的可靠性,并標(biāo)記在樹的分枝上。最后通過(guò)Treeview V32軟件對(duì)樹進(jìn)行編輯處理后導(dǎo)出為圖片用PPT編輯(Page 2013)。

2 結(jié)果與分析

2.1 形態(tài)學(xué)鑒定

2.1.1 厭味紅菇

Russulanauseosa(Pers.) Fr.

菌蓋:30~31×25~28 mm,碟狀或淺杯狀,近圓形,中部略凹陷,印度紅色,由中部到邊緣逐漸變淺,邊緣淺粉色,表面有黏性;菌褶:彎曲、等長(zhǎng)、淺黃色,靠近菌柄處有少量分叉;菌柄:長(zhǎng)40 mm,直徑10 mm,空心,圓柱形,乳白色,基部杵狀,肉質(zhì)薄而易碎(見圖1.a),本實(shí)驗(yàn)研究標(biāo)本:MYH201310045。

生長(zhǎng)環(huán)境:生于針葉林地上。

該種主要分布于吉林、云南、四川等省。

2.1.2 美紅菇

RussulapuellarisFr.

菌蓋:45 mm×45 mm,中部略下凹,后平展,淺粉紅色,有粘性;菌褶:較密、等長(zhǎng),白色,干后乳黃色;菌柄:長(zhǎng)7 cm,直徑:7 mm,圓柱形,直生,實(shí)心至中空,無(wú)附屬物,海綿質(zhì),與菌褶同色,基部杵狀(見圖1.b、圖1.c),本實(shí)驗(yàn)研究標(biāo)本:XJ2013100413。

生長(zhǎng)環(huán)境:生于混交林地上。

該種主要分布于中國(guó)廣東、海南、四川、貴州、遼寧、吉林、江蘇、江西、云南、西藏、河南、湖南、陜西,歐洲、北美洲等地。

2.1.3 鱗蓋紅菇

RussulalepidaFr.

菌蓋:42~51 mm×37~45 mm,扁半球形,猩紅色,無(wú)粘性。菌肉白色,質(zhì)厚,味道鮮美;菌褶:密、等長(zhǎng),白色;菌柄:長(zhǎng)33~45 mm,直徑8~11 mm,圓柱形,實(shí)心,白色略帶紫紅色幼時(shí)緊密,老后松軟(見圖1.d),本實(shí)驗(yàn)研究標(biāo)本:XJ20131004125。

生長(zhǎng)環(huán)境:生于混交林地上。

該種在亞洲、歐洲和北美洲均有分布,均有分布我國(guó)湖北、湖南、廣東、河北、遼寧、吉林、黑龍江、四川、貴州、云南、江蘇、福建、江西、河南、廣西、西藏 、陜西、甘肅等省。

2.1.4 菱紅菇

RussulavescaFr.

菌蓋:62 mm×71 mm,平展,有細(xì)微皺紋或近光滑,淺棕色,邊緣有皺褶;菌褶:密、有分叉,延生,白色;菌柄:長(zhǎng)50 mm,直徑11 mm,圓柱形,內(nèi)海綿質(zhì),靠近菌褶部分與菌褶同色,往下變黃或褐色,基部杵狀。氣味有淡淡的甜味(見圖1.e、圖1.f),本實(shí)驗(yàn)研究標(biāo)本:XJ2013100418。

生長(zhǎng)環(huán)境:生于混交林地上。

該種在亞洲、歐洲和北美洲均有分布,均有分布我國(guó)四川、貴州、云南、遼寧、西藏、新疆、江蘇、福建、江西、湖南、廣東、廣西、海南,以及香港、臺(tái)灣等地。

2.1.5 綠菇

Russulavirescens(Schaeff.) Fr.

菌蓋:74 mm×80 mm,質(zhì)地堅(jiān)實(shí),綠色,初呈球形,長(zhǎng)至后來(lái)呈扁半球形,成熟時(shí)中央稍下凹,淺綠色至灰綠色,表皮往往斑狀龜裂,形成卷毛或小碎片,老時(shí)邊緣有條紋。菌肉白色,味道溫和,無(wú)異味。菌褶:較密、等長(zhǎng),直生,具橫脈,象牙白;菌柄:長(zhǎng)45 mm,直徑16 mm,圓柱形,短而粗,乳白色,基部圓頭狀,中實(shí)或內(nèi)部松軟(見圖1.g、圖1.h),本實(shí)驗(yàn)研究標(biāo)本:XJ2013100520。

生長(zhǎng)環(huán)境:生于混交林地上。

該種在亞洲、歐洲和北美洲均有分布,分布于我國(guó)的西藏、陜西、甘肅、內(nèi)蒙古、遼寧、吉林、黑龍江、四川、貴州、云南、江蘇、浙江、安徽、江西、山東、河南、湖北、湖南、廣東、海南,以及香港、臺(tái)灣等地。

2.1.6RussulafaustianaSarnari

菌蓋:38 mm×40 mm,中間凹陷扁平,外有菌褶向上翻;菌褶:較密、不等長(zhǎng),貼生,有時(shí)分叉,白色;菌柄:長(zhǎng)38 mm,直徑12 mm,圓柱形,乳白色,筆直,基部杵狀(見圖1.i、圖1.j),本實(shí)驗(yàn)研究標(biāo)本:XJ2013100408。

生長(zhǎng)環(huán)境:生于混交林地上。

該種在挪威有記錄,而未在中國(guó)發(fā)現(xiàn)有記錄。

2.1.7 粉柄紅菇

RussulafarinipesRomell

菌蓋:18 ~55 mm×14~46 mm,扁球形,肉質(zhì)硬而有彈性,稻草或奶油黃色,顏色較均勻;菌褶:稀、不等長(zhǎng),土著白;菌褶:向邊緣變薄,白色或淡奶油色;菌柄:長(zhǎng)10~35 mm,直徑6~11 mm,圓柱形,實(shí)心,堅(jiān)硬,土著白,直生或稍彎曲或紡錘型,基部圓形,味辛辣(見圖1.k、圖1.l),本實(shí)驗(yàn)研究標(biāo)本:XJ2013100498。

生長(zhǎng)環(huán)境:生于混交林地上。

該種在亞洲、歐洲和北美洲均有分布,我國(guó)主要集中在吉林、海南、云南、甘肅、江蘇、湖南、廣東、廣西、等省。

2.2 分子系統(tǒng)學(xué)

基于本研究采集的野生紅菇ITSrDNA基因序列排序由508個(gè)堿基組成,其中214個(gè)堿基有簡(jiǎn)約信息位點(diǎn),183個(gè)堿基恒定不變,91個(gè)堿基無(wú)簡(jiǎn)約信息位點(diǎn)。簡(jiǎn)約法分析生成5000棵樹,Kishino-Hasegawa 測(cè)試顯示各個(gè)聚類樹之間沒(méi)有顯著差異,其中一棵如圖2所示(最佳聚類樹),其樹長(zhǎng)(Tree length)=878;一致性指數(shù)(CI)=0.551;保留指數(shù)(RI)=0.714;復(fù)定指數(shù)(RC)= 0.393;非同源性指數(shù)(HI)=0.449。

分子系統(tǒng)學(xué)分析由34個(gè)種組成,其中包括美紅菇Russulapuellaris、鱗蓋紅菇R.lepida、菱紅菇R.vesca、以及綠菇R.virescens、R.melzeri等種類。由圖2中可知,絕大多數(shù)種都能形成單獨(dú)的一簇,而且自展支持率較高,只有少數(shù)爭(zhēng)議種類還待進(jìn)一步核實(shí)形態(tài)后做進(jìn)一步的說(shuō)明。根據(jù)生物條形碼原則,ITS rDNA可以作為紅菇鑒定的較好的條形碼基因。

該系統(tǒng)發(fā)育樹是以GloeocystidiellumaculeatumSheng H. Wu(菌株:AY061739)為外群用PAUP*軟件構(gòu)建的MP系統(tǒng)樹。Bootstrap值為1000次重復(fù)后高于50%以上的值。由圖2可見MYH2013100345號(hào)標(biāo)本與Russulanauseosa,XJ2013100413號(hào)標(biāo)本與R.puellaris,XJ2013100412號(hào)標(biāo)本5與R.lepida,XJ2013100418號(hào)標(biāo)本與R.vesca,XJ2013100520號(hào)標(biāo)本與R.virescens,XJ2013100408號(hào)標(biāo)本與R..faustiana,以及XJ2013100498號(hào)標(biāo)本與R.farinipes均與各自從GenBank檢索比對(duì)得到的最相似物種以100%的bootstrap值聚在了一起,提示這7種紅菇菌株應(yīng)該各自與其最相似物種為同種的紅菇。這一結(jié)果基于ITS序列的分子水平上支持了本研究的形態(tài)鑒定結(jié)果。

3 結(jié)論與討論

本研究表明,綜合形態(tài)與分子鑒定研究結(jié)果,可以確定采自麻陽(yáng)河自然保護(hù)區(qū)的7個(gè)標(biāo)本:MYH2013100345號(hào)標(biāo)本鑒定為Russula.nauseosa(厭味紅菇),XJ2013100413號(hào)標(biāo)本鑒定為R.puellaris(美紅菇),XJ20131004125號(hào)標(biāo)本鑒定為R.lepida(鱗蓋紅菇),XJ2013100418號(hào)標(biāo)本鑒定為R.vesca(菱紅菇),XJ2013100520號(hào)標(biāo)本鑒定為R.virescens(變綠紅菇),XJ2013100408號(hào)標(biāo)本鑒定為R.faustiana,以及XJ2013100498號(hào)標(biāo)本鑒定為R.farinipes(粉柄紅菇)。

紅菇在麻陽(yáng)河自然保護(hù)區(qū)分布廣泛,具有極高的多樣性,是麻陽(yáng)河野生大型真菌的優(yōu)勢(shì)屬。但由于其子實(shí)體形態(tài)特征的分類鑒定不僅受生長(zhǎng)環(huán)境、生長(zhǎng)時(shí)期等客觀因素的影響,而且形態(tài)性狀的選取和判斷都受主觀因素的影響也較大,因而基于形態(tài)性狀特征對(duì)大型真菌進(jìn)行分類鑒定總是有一定的局限性,特別是對(duì)于一些形態(tài)特征極為相近的種,僅憑形態(tài)極難準(zhǔn)確區(qū)分鑒定到種一級(jí)。而真菌的ITS rDNA區(qū)段的保守性基本上表現(xiàn)為種內(nèi)相對(duì)一致、種間差異比較明顯,這一特點(diǎn)使得ITS區(qū)段不僅適用于屬內(nèi)物種間或種內(nèi)差異較明顯的菌群間的系統(tǒng)發(fā)育關(guān)系分析,而且非常適合于真菌物種的分子鑒定[20]。郭亮[21]等利用形態(tài)學(xué)和ITS測(cè)序相結(jié)合的方法鑒定了湖南松乳菇。Zahhoor[22]等利用形態(tài)學(xué)和測(cè)序方法鑒定了Russulaformula和Russulapostina,。所以,本研究在對(duì)麻陽(yáng)河自然保護(hù)區(qū)的紅菇及其它真菌資源調(diào)查的基礎(chǔ)上,充分利用ITS快捷、方便、科學(xué)的特點(diǎn)[23],借助GenBank等生物數(shù)據(jù)庫(kù)的大量信息,從分子水平上為這些紅菇真菌資源的準(zhǔn)確分類、真菌類群的系統(tǒng)發(fā)育提供了科學(xué)依據(jù)。

This study provided a clue for protection and utilization ofRussulaspp. inMayang River reserve and Guizhou Province. Seven species ofRussulaspp. were collected from the Mayang River Nature Reserve in Guizhou Province, China. These species were identified as theR.nauseosa、R.puellaris、R.lepida、R.vesca、R.virescensandR.faustianabased on morphology and phylogenetic analysis of ITS rDNA sequences.
