載體介導(dǎo)shRNA對(duì)胃癌細(xì)胞株MKN45中C—met的抑制作用

【摘要】 目的 探討載體介導(dǎo)短發(fā)夾核糖核酸(shRNA)對(duì)胃癌細(xì)胞株MKN45中原癌基因C-met的抑制作用。方法 9株胃癌細(xì)胞株分為轉(zhuǎn)染組、陰性對(duì)照組和空白對(duì)照組, 各3侏。通過(guò)實(shí)時(shí)熒光定量逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)技術(shù), 觀察轉(zhuǎn)染24 h和48 h后, 原癌基因C-met的信使核糖核酸(mRNA)的CT值的變化;通過(guò)免疫印跡試驗(yàn)(Western blot), 觀察轉(zhuǎn)染后C-met的蛋白變化;通過(guò)四氮唑鹽MTT比色法, 計(jì)算細(xì)胞存活率。結(jié)果 載體介導(dǎo)的C-met shRNA轉(zhuǎn)染24 h和48 h后, RT-PCR結(jié)果顯示, C-met的mRNA抑制率分別為56%和88%;與空白對(duì)照組比較, C-met的蛋白表達(dá)水平明顯下降, 轉(zhuǎn)染后24 h [(0.812±0.105) VS (0.640±0.011)]、48 h[(0.811±0.103) VS (0.331±0.009)], 差異均有統(tǒng)計(jì)學(xué)意義(P<0.05);MTT比色法顯示, 細(xì)胞存活率均有明顯下降, 轉(zhuǎn)染后24 h為65.9%, 48 h為59.6%。結(jié)論 載體介導(dǎo)的C-met shRNA不僅可降低胃癌細(xì)胞株MKN45中原癌基因C-met的mRNA及其蛋白表達(dá)水平, 而且對(duì)癌細(xì)胞的存活有明顯的抑制作用。

【Abstract】 Objective To investigate inhibiting effect by vector-mediated short hairpin RNA (shRNA) on proto-oncogene C-met in stomach cancer cell strain MKN45. Methods A total of 9 stomach cancer cell strains were divided into transfection group, negative control group and blank control group, with 3 strains in each group. Real-time fluorescent quantitative reverse transcription-polymerase chain reaction (RT-PCR) was applied to observe changes of CT value of messenger ribonucleic acid (mRNA) in proto-oncogene C-met after 24 and 48 h of transfection. Western blot was used to observe changes of protein in C-met after transfection. Tetrazolium-based colorimetric assay (MTT) was applied to calculate cell survival rate. Results In 24 and 48 h after vector-mediated mRNA shRNA transfection, RT-PCR showed mRNA inhibition rates by C-met were respectively 56% and 88%. Comparing with the blank control group, C-met had obviously reduce protein expression level, as [(0.812±0.105) VS (0.640±0.011)] in 24 h after transfection and [(0.811±0.103) VS (0.331±0.009)] in 48 h after transfection. Their differences all had statistical significance (P<0.05), MTT showed obviously decreased cell survival rate, as 65.9% in 24 h after transfection and 59.6% in 48 h after transfection. Conclusion Vector-mediated C-met shRNA can not only reduce mRNA and its protein expression in proto-oncogene C-met of stomach cancer cell strain MKN45, and also show remarkable inhibiting effect for survival of cancer cell.

據(jù)Roder[1]報(bào)道, 我國(guó)為胃癌的高發(fā)地區(qū)。有研究發(fā)現(xiàn)[2], RNA干擾是人為引入與內(nèi)源靶基因具有相同序列雙鏈RNA, 誘導(dǎo)內(nèi)源靶基因的mRNA降解, 達(dá)到阻止基因表達(dá)的目的。現(xiàn)將研究結(jié)果報(bào)告如下。

1 材料與方法

1. 1 材料 胃癌細(xì)胞株MKN45購(gòu)自上海市消化疾病研究所(共9株);實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)試劑盒購(gòu)自美國(guó)Promega公司;小鼠抗人β-肌動(dòng)蛋白(β-actin)一抗、小鼠抗人C-met一抗、辣根過(guò)氧化物物酶標(biāo)記兔抗小鼠二抗購(gòu)自美國(guó)Santa Cruz公司; C-met shRNA和陰性對(duì)照購(gòu)自美國(guó)OPEN-BIOSYSTEMS公司。

1. 2 方法 實(shí)驗(yàn)分組:轉(zhuǎn)染組(轉(zhuǎn)染shRNA)、陰性對(duì)照組(轉(zhuǎn)染錯(cuò)義序列)和空白對(duì)照組(未轉(zhuǎn)染shRNA), 各3株。細(xì)胞轉(zhuǎn)染, RT-PCR檢測(cè)各組的C-met的 mRNA表達(dá), 讀取每孔的CT值[3, 4], 計(jì)算各分組目的基因相對(duì)定量值。Western blot法檢測(cè)C-met的蛋白表達(dá):收集各分組細(xì)胞, 加入稀釋過(guò)的抗體(小鼠抗人β-actin一抗1∶150、小鼠抗人C-met一抗1:150)4℃孵育過(guò)夜, 漂洗后加入辣根過(guò)氧化物物酶標(biāo)記兔抗小鼠二抗, 化學(xué)發(fā)光法顯色, 用目的蛋白C-met A值與內(nèi)參β-actin A值的比值分析[5]。MTT比色試驗(yàn)法檢測(cè)490 nm的吸收值(A), 計(jì)算細(xì)胞存活率[6, 7]。

1. 3 統(tǒng)計(jì)學(xué)方法 采用SPSS19.0統(tǒng)計(jì)學(xué)軟件進(jìn)行統(tǒng)計(jì)分析。計(jì)量資料以均數(shù)±標(biāo)準(zhǔn)差( x-±s)表示, 采用t檢驗(yàn)。P<0.05表示差異具有統(tǒng)計(jì)學(xué)意義。

2 結(jié)果

2. 1 RT-PCR檢測(cè)C-met的mRNA表達(dá)情況 RT-PCR結(jié)果顯示, C-met的mRNA抑制率分別為56%和88%;RT-PCR檢測(cè)各組C-met的mRNA表達(dá)水平見(jiàn)表1。

2. 2 Western blot檢測(cè)C-met 的蛋白表達(dá)水平 與空白對(duì)照組相比, 轉(zhuǎn)染C-met shRNA 24 h MKN45細(xì)胞中的C-met蛋白含量(0.640±0.011)和48 h MKN45細(xì)胞中的C-met蛋白含量(0.331±0.009)顯著降低, 差異均有統(tǒng)計(jì)學(xué)意義(P<0.05);而陰性對(duì)照組C-met蛋白含量分別為(0.809±0.101)、(0.808±0.104)和空白對(duì)照組C-met蛋白(0.812±0.105)、(0.811±0.1033)無(wú)明顯抑制, 差異無(wú)統(tǒng)計(jì)學(xué)意義(P>0.05)。

2. 3 MTT試驗(yàn)結(jié)果 轉(zhuǎn)染后MTT法檢測(cè)胃癌細(xì)胞株MKN45的細(xì)胞存活率, C-met shRNA轉(zhuǎn)染24 h后存活率為65.9%、轉(zhuǎn)染48 h后存活率為59.6%, 均低于陰性對(duì)照組的96.3%、98.4%。

3 討論

鑒于原癌基因C-met在胃癌的發(fā)生和發(fā)展中的重要作用及RNAi技術(shù)在腫瘤基因治療中的巨大潛在價(jià)值。RNAi技術(shù)可有效沉默胃癌細(xì)胞株中的原癌基因C-met的表達(dá), 誘導(dǎo)腫瘤細(xì)胞的凋亡, 但研究多數(shù)采用的是siRNA或者miRNA技術(shù)。本研究應(yīng)用shRNA技術(shù), 通過(guò)沉默原基基因C-met的表達(dá), 降低腫瘤細(xì)胞的存活率, 誘導(dǎo)腫瘤細(xì)胞凋亡。

本研究采用的C-met shRNA, 簡(jiǎn)單、易操作, 而且該C-met shRNA轉(zhuǎn)染高, 其設(shè)計(jì)的沉默序列可保證有效、有針對(duì)性地沉默腫瘤細(xì)胞中的C-met的表達(dá)。本研究通過(guò)RT-PCR、Western blot和MTT實(shí)驗(yàn)結(jié)果可知, C-met shRNA成功沉默原癌基因C-met, 致使其mRNA及蛋白表達(dá)明顯下降, 并使腫瘤細(xì)胞存活率下降, 成功誘導(dǎo)胃癌細(xì)胞凋亡;比較轉(zhuǎn)染C-met shRNA 24 h及48 h結(jié)果可知, 隨著轉(zhuǎn)染時(shí)間的增長(zhǎng), C-met shRNA沉默作用也增強(qiáng);而比較空白對(duì)照組和陰性對(duì)照組結(jié)果可知, 介導(dǎo)轉(zhuǎn)染細(xì)胞的載體對(duì)原癌基因C-met無(wú)直接抑制作用, 關(guān)鍵在于C-met shRNA的直接抑制作用。
