血紅蛋白F與血紅蛋白A2之間的交叉互作

目的我們曾經(jīng)用淀粉-瓊脂混合凝膠電泳證明成人血紅蛋白A1與A2發(fā)生交叉互作，當時未對胎兒血紅蛋白F進行研究?，F(xiàn)在，再用這種方法研究血紅蛋白F與血紅蛋白A2是否也有相互作用。方法由臍帶血分離紅細胞，再用淀粉-瓊脂糖混合凝膠電泳制備純的血紅蛋白F，然后讓血紅蛋白F穿過成人紅細胞溶血液，觀察穿過溶血液后血紅蛋白A2區(qū)帶變形情況。結(jié)果血紅蛋白F穿過成人紅細胞溶血液中的血紅蛋白A2時，后者變形，與游離血紅蛋白F相比，進入溶血液中的血紅蛋白F泳速稍慢。結(jié)論血紅蛋白F與血紅蛋白A2也能發(fā)生交叉互作。

本文所使用的技術(shù)屬于交叉電泳范疇，在這方面日本學(xué)者中村正二郎曾專門著書[1]介紹他們利用紙上電泳和瓊脂電泳研究血清蛋白及免疫球蛋白的研究成果。我們所使用的支持體為淀粉-瓊脂糖混合凝膠，不同于紙上電泳和瓊脂電泳，研究對象也不一樣，這里是血紅蛋白。用本法發(fā)現(xiàn)成人血紅蛋白A1與A2之間發(fā)生交叉互作[2-4]。本文的研究內(nèi)容是胎兒血紅蛋白F與成人血紅蛋白A2之間是否存在交叉互作問題。

1 材料與方法

2 結(jié) 果

血紅蛋白F穿過成人紅細胞溶血液后的結(jié)果見圖1，由此圖可以看出，血紅蛋白F穿過成人紅細胞溶血液中的血紅蛋白A2時，后者變形（參見箭↓頭處），與游離血紅蛋白F相比，進入溶血液中的血紅蛋白F泳速稍慢。

3 討 論

血紅蛋白F穿過血紅蛋白A2時，A2區(qū)帶也變形，說明二者也發(fā)生交叉互作。此時，血紅蛋白F泳速減慢，說明穿過時二者曾一度結(jié)合并很快分開，它們的反應(yīng)式如下：

穿過實驗的安排是，將慢泳成分（血紅蛋白A2）放在前面，快泳成分（血紅蛋白F）放在后邊，電泳過程中F趕上并進入A2時反應(yīng)向右進行，出現(xiàn)一過性交叉互作產(chǎn)物。繼續(xù)電泳，F(xiàn)超過A2時反應(yīng)向左進行，又恢復(fù)為HbF和HbA2，但留下了血紅蛋白A2變形的痕跡。

我們以前發(fā)現(xiàn)的成人HbA1和HbA2交叉互作[2-4]也是同樣道理，那時的反應(yīng)式如下：

現(xiàn)在看來，這里是兩個血紅蛋白二聚體一過性結(jié)合成四聚體，然后再分開，留下了結(jié)合過的痕跡，讓我們發(fā)現(xiàn)了它們的交叉互作。

以上是人體內(nèi)部幾種血紅蛋白之間的交叉互作，人體血紅蛋白與各種動物血紅蛋白之間有無交叉互作，各種動物血紅蛋白之間有無交叉互作，都是需要進一步研究有趣課題。在這些方面，我們已經(jīng)做了一些工作[5-15]，等待再深入。

Cross Interaction Between HbF and HbA2

ObjectiveWe have demonstrate that adult hemoglobin HbA1and HbA2can cross interact each other by starch-agarose mixed gel electrophoresis, but the fetal hemoglobin HbF was not studied at that time. At present, we use this method to study whether the interaction between HbF and HbA2occur.MethodsSeparating red blood cells from the umbilical cord, then preparation pure hemoglobin F from the starch-agarose electrophoresis gel electrophoresis, and let hemoglobin F dissolve blood through adult red blood cells was observed after dissolving blood hemoglobin A2through the deformation zone.ResultsThe dissolution of erythrocyte hemoglobin F in the blood through the adult hemoglobin A2, which is deformed as compared with the free hemoglobin F, hemoglobin in the blood solution into the F swimming speed slower.ConclusionThe results showed that the cross interaction was occurred as well between HbF and HbA2.
