摘 要 目的:觀察殼聚糖對大鼠消化性潰瘍愈合的影響,探討其抗?jié)兊目赡軝C制。方法:采用冰乙酸胃竇部漿膜下注射法制備大鼠消化性潰瘍模型。70只實驗大鼠被隨機分成7組,即假手術(shù)對照組、模型對照組、泮托拉唑治療組、硫糖鋁混懸液治療組及3個不同劑量殼聚糖治療組( 0.25 g/kg,0.5 g/kg,1.0 g/kg)。按Guth標(biāo)準(zhǔn)記分判定潰瘍指數(shù)并計算潰瘍抑制率;HE染色觀察組織結(jié)構(gòu)。采用羥胺法測定超氧化物岐化酶(SOD),DTNB法測定谷胱甘肽-過氧化物酶(GSH-Px),TAB法測定丙二醛(MDA)。結(jié)果:殼聚糖治療組與模型對照組比較,潰瘍指數(shù)明顯降低,潰瘍抑制率升高(P < 0.01);1.0 g/kg殼聚糖治療組大鼠血清SOD、MDA、GSH-Px值與模型對照組均有顯著性差異(P<0.05); 0.25 g/kg殼聚糖治療組及0.5 g/kg殼聚糖治療組大鼠血清MDA值與模型對照組比較均有顯著性差異(P< 0.05),但SOD、GSH-Px值與模型對照組比較均無顯著性差異。結(jié)論:殼聚糖可促進(jìn)冰乙酸潰瘍大鼠的潰瘍愈合,并呈一定的量效關(guān)系。其促進(jìn)潰瘍愈合作用可能與抗氧化機制有關(guān)。

Effect of chitosan on the serum levels of MDA, SOD, and GSH-Px in rats with gastric ulcer

ABSTRACT Aim:
To observe the effect of chitosan on the healing of gastric ulcer and explore the possible mechanisms for its action. Methods: Glacial acetic acid was subserosally injected to the gastric antrum of rats to establish the animal model of gastric ulcer. Seventy rats were randomly divided into 7 groups: sham-operated group, model group, pantoprazole-treated group, sucralfate-treated group, and three chitosan-treated groups at low, moderate and high doses (0.25, 0.5, and 1.0 g/kg). The ulcer index was evaluated by Guth method and the inhibition ratio of ulcer formation was calculated. The morphological changes were observed by HE staining. The levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px),and malondialdehyde (MDA) were determined by hydroxylamine method, DTNB method and TAB method respectively. Results: The ulcer index was significantly decreased and the inhibition ratio of ulcer formation was significantly increased in chitosan-treated groups compared with model group (P<0.01). The serum levels of MDA, SOD and GSH-Px in chitosan 1.0 g/kg group had significant difference compared with those of model group (P<0.05). The serum level of MDA in chitosan 0.25 g/kg group and 0.5 g/kg group were significantly different with that of model group (P<0.05), but there was no significant difference in the serum levels of SOD and GSH-Px among chitosan 0.25 g/kg group and 0.5 g/kg groups and model group. Conclusion: Chitosan may improve the healing of gastric ulcer induced by glacial acetic acid in rats. The effect was in a dose-dependent manner. The action mechanism of chitosan may be related with its antioxidant activities.

消化性潰瘍是一類常見病、多發(fā)病。隨著藥物治療的進(jìn)展,消化性潰瘍的愈合率也日趨提高,但不能避免復(fù)發(fā)。
殼聚糖由甲殼素加工而成,對人體安全無毒[1~3],可生物降解,具有良好的生物相容性。殼聚糖的水解產(chǎn)物是表皮細(xì)胞或組織生長必需的生物活性物質(zhì),有利于潰瘍局部肉芽組織和新生血管的形成;殼聚糖分子中含有極性基團(tuán)并帶正電荷,溶解于胃酸,形成黏稠的膠,附著在胃壁上形成一層保護(hù)膜,阻止胃酸和胃蛋白酶對潰瘍面的腐蝕,促進(jìn)潰瘍修復(fù)和愈合。國內(nèi)外動物實驗證實[4~9],殼聚糖有保護(hù)胃黏膜、促進(jìn)胃潰瘍愈合作用。本文對殼聚糖抗?jié)兊目赡軝C制作一研究。
1 材料與方法
1.1 材料
動物:雄性Sprague-Dawley系大鼠,體重190~250 g,共70只,購自西普爾-必凱實驗動物有限公司。
藥物:殼聚糖,脫乙酰度92%,華東理工大學(xué)提供;泮托拉唑鈉腸溶片,沈陽東宇藥業(yè)有限公司生產(chǎn);硫糖鋁混懸液,上海旭東海普藥業(yè)有限公司生產(chǎn)。
試劑:超氧化物歧化酶(superoxide dismutase,SOD)測試盒、丙二醛(malonaldehyde,MDA) 測試盒及谷肽甘肽-過氧化物酶 (glutathione peroxidase ,GSH-Px) 測試盒,均購自南京建成生物工程研究所。
