GATA3蛋白在小鼠胚胎發(fā)育中的表達(dá)研究

GATA3蛋白在小鼠胚胎發(fā)育中的表達(dá)研究

為進(jìn)一步探討GATA3在蛋白水平上的時(shí)空表達(dá)模式,應(yīng)用熒光免疫組織化學(xué)技術(shù)對(duì)其在小鼠胚胎發(fā)育各時(shí)期的表達(dá)進(jìn)行了研究.結(jié)果顯示:從E11.5到E17.5時(shí)期,其表達(dá)在神經(jīng)系統(tǒng)(腦橋和脊髓)中隨胎齡的增大而減弱;在感官系統(tǒng)(犁鼻器)中隨胎齡的增大而位置有所遷移;在泌尿系統(tǒng)(腎臟)中隨胎齡的增大而增強(qiáng).

0 引言

真核生物中,GATA家族由能結(jié)合特異DNA序列(A/T GATA A/G)的鋅指狀結(jié)構(gòu)轉(zhuǎn)錄因子組成,與個(gè)體發(fā)育和細(xì)胞分化等息息相關(guān).其成員之一GATA3,在中樞神經(jīng)系統(tǒng)[1]、內(nèi)耳[2]、交感神經(jīng)系統(tǒng)[3]、免疫系統(tǒng)[4]、造血系統(tǒng)[5]、皮膚和毛囊細(xì)胞[6]、腎管[7]、晶狀體[8]、乳腺祖細(xì)胞[9]、乳腺癌轉(zhuǎn)移[10]等方面扮演了非常關(guān)鍵的角色.此外,GATA3單倍劑量不足會(huì)導(dǎo)致常染色體顯性遺傳病甲狀旁腺功能減退-感音神經(jīng)性耳聾-腎發(fā)育不良(HDR)綜合征的發(fā)病[11-12].目前為止,關(guān)于GATA3的功能和作用尚未被完全闡述清楚.

表達(dá)預(yù)示著功能.現(xiàn)有對(duì)于GATA3的表達(dá)研究?jī)H停留在mRNA水平[13-15],并未將其在各組織器官的時(shí)空表達(dá)模式闡述完全[16-17].因此,本文應(yīng)用熒光免疫組織化學(xué)技術(shù)系統(tǒng)地研究了GATA3蛋白在小鼠胚胎發(fā)育中的時(shí)間空間表達(dá).

1 材料和方法

1.1 材料

1.1.1 供試動(dòng)物

C57BL/6健康小鼠,6周齡雌鼠、6周齡雄鼠(購(gòu)于杭州師范大學(xué)實(shí)驗(yàn)動(dòng)物研究中心).在室溫22 ℃、控光(12 h晝夜節(jié)律)條件下按常規(guī)飼養(yǎng).所購(gòu)小鼠至少適應(yīng)環(huán)境1周后方用于實(shí)驗(yàn).

1.1.2 小鼠胚胎的獲取

第一天將雄性小鼠和雌性小鼠按1∶3比例合籠,次日上午9時(shí)觀察雌性小鼠陰栓情況,陰栓陽(yáng)性小鼠視為交配成功,另外分籠飼養(yǎng)并標(biāo)明交配日期,從交配成功日起,計(jì)算受孕為0.5 d,依據(jù)實(shí)驗(yàn)要求在不同時(shí)間段處理胚胎.

1.1.3 主要試劑

OCT冰凍切片包埋劑,磷酸緩沖溶液(PBS),4%多聚甲醛溶液(PFA),10%,20%和30%蔗糖溶液,0.2% PBST,牛血清白蛋白(BSA),一抗Anti-Gata3(abcam,ab199428),二抗Anti-Rabbit(568 nm,life technologies,A10042).

1.2 冰凍切片

分別于胚胎發(fā)育至11.5,12.5,13.5,14.5,15.5,16.5,17.5和18.5 d時(shí),采取斷頸方法處死孕鼠取出胚胎,將胚胎置于PBS(4 ℃預(yù)冷)中剝除胎衣,再轉(zhuǎn)移至4%多聚甲醛溶液,固定(注:15.5 d及以后胎齡的胚胎需頭身分離分別固定).然后用0.1 mol/L PBS 15 min 洗3次,再依次加入10%,20%,30%蔗糖溶液梯度脫水,直至組織降至液體底部,徹底脫水防止冰晶形成,接著OCT包埋后于-80 ℃冰凍保存.用冰凍切片機(jī)切片,厚度為20 μm.

1.3 熒光免疫組織化學(xué)

切好的組織片放入1× PBS中洗2次,每次15 min;接著放入含0.2%Triton×-100的PBST中洗2次,每次15 min;用含2%BSA的封閉液,室溫封閉1 h;加入按比例稀釋的一抗,4 ℃過夜;片子取出后放入PBST中洗4次,每次10 min;再加入按比例稀釋的二抗,室溫孵育1.5 h;在1×PBST洗2次,每次5 min;然后加入DAPI,室溫 5 min;在1×PBS中洗2次,每次5 min,封片拍照.抗體為Anti-Gata3(稀釋比例1∶500),Anti-Rabbit(稀釋比例1∶1 000).所有實(shí)驗(yàn)均重復(fù)3次.

2 結(jié)果

2.1 GATA3在腦橋的表達(dá)情況

腦橋是中樞神經(jīng)系統(tǒng)中小腦和延髓之間的重要連接部位.從E11.5到E13.5時(shí)期,GATA3在腦橋部位均有表達(dá)(圖1A,C,E),且表達(dá)強(qiáng)度隨時(shí)間增加呈現(xiàn)明顯的減弱趨勢(shì)(圖1B,D,F(xiàn)).當(dāng)E13.5時(shí)呈明顯的點(diǎn)狀散在分布(圖1F),之后表達(dá)消失.

2.2 GATA3在脊髓的表達(dá)情況

在E12.5,E15.5,E17.5時(shí),GATA3在脊髓橫斷切片的腹側(cè)部均有表達(dá),且表現(xiàn)出明顯的逐漸減少趨勢(shì)(圖2).

2.3 GATA3在背根神經(jīng)節(jié)的表達(dá)情況

矢狀面切片顯示E13.5時(shí),GATA3在背根神經(jīng)節(jié)表達(dá)明顯(圖3),之后其它時(shí)期未檢出.

2.4 GATA3在犁鼻器的表達(dá)情況

從E12.5到E15.5,GATA3在犁鼻器的呼吸上皮層表達(dá)(圖4),并隨時(shí)間增加呈明顯的從兩邊向中間聚攏趨勢(shì)(圖4B,D,E,F(xiàn)).

2.5 GATA3在舌的表達(dá)情況

當(dāng)E13.5時(shí),GATA3在舌部表達(dá)(圖5),呈規(guī)律的條紋狀,其在E11.5表達(dá)不明顯,之后的時(shí)期表達(dá)消失.

2.6 GATA3在腎的表達(dá)情況

從E12.5到E17.5, GATA3完全且局限于腎小管上皮部位,并隨時(shí)間增加呈現(xiàn)遞增趨勢(shì)(圖6).

3 討論

已有的原位雜交實(shí)驗(yàn)結(jié)果[1,15]顯示:中樞神經(jīng)系統(tǒng)中,GATA3在腦橋、脊髓和背根神經(jīng)節(jié)的表達(dá)都始于E11.5;在腦橋的表達(dá)于E12.5時(shí)減少,E14.5時(shí)限制更加嚴(yán)重;在脊髓的表達(dá)于E12.5時(shí)只限于腹側(cè);在背根神經(jīng)節(jié)的表達(dá)于E12.5時(shí)消失.另外,E14.5時(shí),在犁鼻器檢測(cè)到Gata3mRNA,但缺乏對(duì)其時(shí)空表達(dá)變化的研究.犁鼻器(VON)在鼠類的社會(huì)行為中發(fā)揮著化學(xué)信號(hào)調(diào)節(jié)作用,特別是對(duì)攻擊命令等[18].

有別與前述研究,筆者分析了GATA3在蛋白水平上的時(shí)空表達(dá),結(jié)果發(fā)現(xiàn):在腦橋的表達(dá)從E11.5到E13.5,且呈強(qiáng)度減弱趨勢(shì),E14.5之后的表達(dá)消失(圖1);在脊髓,從E12.5到E17.5時(shí)期,表達(dá)持續(xù)存在于腹側(cè),呈逐漸減少趨勢(shì)(圖2);背根神經(jīng)節(jié),E13.5時(shí)表達(dá)明顯,之后其他時(shí)期未檢出(圖3);在犁鼻器部位,從E12.5到E15.5時(shí)期表達(dá)在呼吸上皮層,且呈明顯的從兩邊向中間聚攏趨勢(shì)(圖4),預(yù)示著GATA3可能在呼吸系統(tǒng)發(fā)育中發(fā)揮重要功能,需要進(jìn)一步的探討.GATA3在舌頭部位的表達(dá)結(jié)果顯示E13.5時(shí)表達(dá)明顯(圖5),其作用尚需進(jìn)一步探究.

此外,關(guān)于腎功能缺陷與GATA3基因突變的關(guān)系,之前曾有相左的病例報(bào)道[19-20],雖然有對(duì)相關(guān)機(jī)制的探討[21-22],但還未闡述清楚.本文的實(shí)驗(yàn)結(jié)果顯示:GATA3在腎臟的表達(dá)從E12.5到E17.5呈現(xiàn)遞增趨勢(shì),且隨腎小管的增大而增大,完全且局限于各個(gè)獨(dú)立的腎小管上皮部位,這對(duì)將來其功能的機(jī)制探討有參考價(jià)值.

綜上所述,本研究更加系統(tǒng)地探討了GATA3在蛋白水平上的時(shí)空表達(dá)模式,從小鼠胚胎E11.5到E17.5時(shí)期,在神經(jīng)系統(tǒng)(腦橋、脊髓、背根神經(jīng)節(jié))、感官系統(tǒng)(犁鼻器、舌)以及泌尿系統(tǒng)(腎臟),都有新發(fā)現(xiàn).本文研究結(jié)果對(duì)GATA3在蛋白水平上的表達(dá)進(jìn)行了深入闡述,也是對(duì)之前相關(guān)數(shù)據(jù)的完善.

To further discuss the temporal and spatial expression pattern of GATA3 on protein level, immunofluorescence histochemical technique is applied to study the expression of GATA3 in mouse embryonic development. The results show that from E11.5 to E17.5 period, the expression of GATA3 in nervous system (pons and spinal cord) decreases with the increase of gestational age, migrates with the increase of gestational age in sensory system (vomeronasal organ), and increases with the increase of gestational age in urinary system (kidney).
