新型甾體N,N-二甲基縮氨硫腙類化合物的合成及其抗腫瘤活性*

摘要:以乙醇(或甲醇為溶劑),冰醋酸為催化劑,去氫表雄酮、孕烯醇酮、雌酮和3,5-二羥基-6-甲?;?B-降膽甾烷分別與N,N-二甲基硫代氨基脲經(jīng)縮合反應(yīng),合成了4個(gè)新型的具有不同甾核結(jié)構(gòu)特征的甾體N,N-二甲基縮氨硫腙類化合物(1~4),其結(jié)構(gòu)經(jīng)1H NMR,13C NMR,IR和HR-EI-MS表征。采用MTT法測(cè)定了1~4對(duì)肝癌細(xì)胞(HepG)、人鼻咽癌細(xì)胞(CNE-2)和人腎上皮細(xì)胞(HEK-293T)的體外抑制細(xì)胞生長增殖活性。結(jié)果表明:1~3對(duì)HepG和CNE-2具有明顯的抑制活性。

近來,人們發(fā)現(xiàn)在甾體化合物中引入氮,硫原子或與含氮,硫等雜原子的藥效基團(tuán)拼合后,會(huì)產(chǎn)生獨(dú)特的生物活性,用于治療不同的疾?。?-6]。腙類化合物是有機(jī)化學(xué)家關(guān)注的焦點(diǎn)[7-8],許多結(jié)構(gòu)獨(dú)特的甾體腙類化合物已相繼被合成,并且許多具有良好的抗腫瘤、抗菌及抗病毒活性[9-11]。其中,縮胺硫腙化合物因其良好的腫瘤細(xì)胞毒性、抗菌活性及抗病毒活性,日益受到藥物學(xué)家關(guān)注。在甾體中引入縮胺硫腙藥效團(tuán)的化合物有很好的鎮(zhèn)痛、抗菌、抗真菌及抗炎活性[12-16]。但至今為止,具有抗腫瘤活性的甾體縮胺硫腙化合物報(bào)道較少。

本課題組曾分別以膽固醇、孕烯醇酮、豆甾醇等為原料,合成了一系列甾體肟類及縮氨硫腙類化合物。體外抗腫瘤活性研究發(fā)現(xiàn)其有較好的抑制腫瘤細(xì)胞生長增殖活性[17-20]。2010年,Huang H等[21]設(shè)計(jì)并合成了一系列結(jié)構(gòu)新穎的稠環(huán)類縮胺硫腙化合物,并深入研究了其抗腫瘤活性和作用機(jī)制。其中部分化合物表現(xiàn)出顯著抑制人腫瘤細(xì)胞生長增殖活性,特別是具有N,N-二甲基縮氨硫腙結(jié)構(gòu)的化合物TSC24(Chart 1)活性最強(qiáng),對(duì)不同腫瘤細(xì)胞株抑制活性的IC50值在2 μmol· L-1~830 μmol·L-1

受此啟發(fā),本文在文獻(xiàn)[21]方法的基礎(chǔ)上,以乙醇(或甲醇為溶劑),冰醋酸為催化劑,去氫表雄酮、孕烯醇酮、雌酮和3,5-二羥基-6-甲?;?B-降膽甾烷分別與N,N-二甲基硫代氨基脲經(jīng)縮合反應(yīng),合成了4個(gè)新型的具有不同甾核結(jié)構(gòu)特征的甾體N,N-二甲基縮氨硫腙化合物(1~4,Scheme 1),其結(jié)構(gòu)經(jīng)1H NMR,13C NMR,IR和HR-EI-MS確證。測(cè)定了1~4對(duì)腫瘤細(xì)胞[肝癌細(xì)胞(HepG)、人鼻咽癌細(xì)胞(CNE-2)和人腎上皮細(xì)胞(HEK-293T)]的生長增殖活性,希望利用甾體化合物滲透細(xì)胞膜的功能,獲得具有更好抗腫瘤活性的甾體縮氨硫腙化合物。

1 實(shí)驗(yàn)部分

1.1儀器與試劑

X6型顯微熔點(diǎn)儀(溫度未校正); Bruker AV-300型超導(dǎo)核磁共振儀(CDCl3為溶劑,TMS為內(nèi)標(biāo)); Thermo Scientific Nicolet IS-10型傅立葉紅外光譜儀(KBr壓片); Agilent 6210 TOFMS型質(zhì)譜儀; MLLTISKAN MK3型分光光度儀。

3,5-二羥基-6-甲?;?B-降膽甾烷參考文獻(xiàn)[19]方法合成;去氫表雄酮、孕烯醇酮,雌酮和N,N-二甲基硫代氨基脲,百靈威科技有限公司;薄層層析板和硅膠,安徽良臣硅源材料有限公司;其余所用試劑均為分析純,溶劑按常規(guī)方法干燥除水。

1.2 1~4的合成通法

在反應(yīng)瓶中加入甾醇0.40 mmol,N,N-二甲基氨基硫脲0.41 mmol和無水乙醇25 mL,于75℃攪拌使其完全溶解;加入冰醋酸0.5 mL,反應(yīng)4 h(TLC跟蹤)。減壓蒸除大部分溶劑后加入蒸餾水20 mL,用乙酸乙酯(3×20 mL)萃取,合并有機(jī)相,依次用飽和碳酸氫鈉溶液(30 mL)和飽和氯化鈉溶液(30 mL)洗滌,無水硫酸鈉干燥;減壓蒸除溶劑后經(jīng)柱層析[洗脫劑:V(石油醚)∶V(乙酸乙酯)=2∶1]純化得白色固體1~3和棕色油狀液體4。

N,N-二甲基-去氫表雄酮-17-縮氨硫腙(1):產(chǎn)率32%,m.p.200℃~202℃;1H NMR δ:0.92(s,3H,18-H),1.04(s,3H,19-H),3.33(s,6H,NCH3),3.48~3.59(m,1H,3-Hα),5.37(d,J=4.8 Hz,1H,6-H),7.87(br s,1H,NH);13C NMR δ:16.8(C18),19.4(C19),20.5(C11),23.5(C15),29.7(C16),31.2(C8),31.3(C7),31.6(C2),33.9(C12),34.0(C10),36.6(NCH3),37.2(C1),42.2(C4),44.5(C13),50.3(C9),53.8(C14),71.6(C3),121.0(C6),141.0(C5),165.8(C17),182.9(C=S); IR ν:3 424,2 925,1 644,1 539,1 464,1 364,1 260,1 118,1 053,908,734 cm-1; HR-EI-MS m/z:Calcd for C22H36N3OS{[M +H]+}390.257 9,found 390.258 6。

N,N-二甲基孕甾-20-縮氨硫腙(2):產(chǎn)率42%,m.p.217℃~218℃;1H NMR δ:0.67(s,3H,18-H),1.02(s,3H,19-H),1.26(s,3H,21-CH3),3.31(s,6H,NCH3)3.48~3.59(m,1H,3-Hα),5.36(d,J=4.8 Hz,1H,6-H);13C NMR δ:13.5(C18),19.4(C19),20.9(C21),21.0(C11),23.7(C15),24.2(C16),29.7(C2),31.6(C8),32.1(C7),36.5(C10),36.5(NCH3),37.3(C1),38.4(C12),42.2(C4),45.2(C13),50.0(C9),56.3(C17),58.0(C14),71.7(C3),121.2(C6),140.8(C5),152.1(C20),183.0(C=S); IR ν:3 424,2 925,1 726,1 504,1 384,1 314,1 254,1 122,1 050,908,726 cm-1; HR-EI-MS m/z:Calcd for C24H40N3OS{[M + H]+} 418.289 2,found 418.290 3。

1.3抗腫瘤活性測(cè)定

以順鉑為陽性對(duì)照,采用MTT法測(cè)定了1~4 對(duì)HepG,CNE-2和HEK-293T的體外生長增殖抑制活性。將對(duì)數(shù)生長期的細(xì)胞分別以(3~4)×104個(gè)/mL的密度接種于96孔板中,每孔接種200 μL,置CO2培養(yǎng)箱中培養(yǎng)24 h。按預(yù)設(shè)的濃度梯度分別加入待測(cè)樣品,每一個(gè)濃度梯度設(shè)3個(gè)平行孔,同時(shí)設(shè)對(duì)照孔和空白孔。保持37℃在CO2培養(yǎng)箱中培養(yǎng)72 h。每孔加入20 μL的MTT(5 mg· mL-1)溶液,再于CO2培養(yǎng)箱中溫育4 h。棄去上清液,加入DMSO 200 μL,混勻10 min,待沉淀完全溶解后,用酶標(biāo)儀在490 nm處測(cè)定溶液的OD值,計(jì)算一定濃度下化合物對(duì)細(xì)胞的抑制率;以抑制率對(duì)藥物濃度作圖,分別求出每個(gè)樣品的IC50值。

2 結(jié)果與討論

2.1合成與表征

從去氫表雄酮、孕烯醇酮和雌酮出發(fā),其17-位或20-位羰基與N,N-二甲基硫代氨基脲發(fā)生縮合反應(yīng),引入N,N-二甲基氨基硫腙基團(tuán),合成了1~3。

1的1H NMR分析表明,δ 3.33處單峰為6個(gè)氫[N(CH3)2,此處與氮原子相連的兩甲基是等位的,故表現(xiàn)為一個(gè)單峰],δ 7.87處的寬單峰是與氮相連氫的吸收峰。13C NMR分析表明,δ 182.9處出現(xiàn)C=S吸收峰,δ 165.8處出現(xiàn)C=N吸收峰,去氫表雄酮17-位羰基碳在δ 220處的吸收峰消失,均表明去氫表雄酮中的17-羰基已與N,N-二甲基硫代氨基脲縮合合成了1。2和3的表征與1類似。

4的1H NMR分析表明,δ 7.21處的吸收峰是與氮相連6-位氫吸收峰,δ 4.10處的吸收峰是與羥基相連3-位氫吸收峰,δ 3.33處的單峰是與氮相連的兩個(gè)甲基的6個(gè)氫吸收峰。13C NMR分析表明,δ 67.1和δ 83.6吸收峰分別是與羥基相連的C3和C5;δ 180.9處吸收峰歸屬C=S,δ 150.7處吸收峰則歸屬C=N,進(jìn)一步結(jié)合質(zhì)譜數(shù)據(jù)可確定4的結(jié)構(gòu)。

2.2抗腫瘤活性

1~4的抗腫瘤活性見表1。由表1可見,1~4對(duì)所測(cè)試的腫瘤細(xì)胞株均具有一定的抑制生長增殖活性,其中1對(duì)CNE-2的IC50為15.4 μmol· L-1,優(yōu)于陽性對(duì)照物順鉑; 2對(duì)HepG的IC50為

10.7 μmol·L-1。有趣的是具有17-縮氨硫腙結(jié)構(gòu)的化合物1和3對(duì)HEK-293T沒有明顯的細(xì)胞毒性,值得進(jìn)一步深入研究。

3 結(jié)論

以具有不同甾核結(jié)構(gòu)特征的甾酮或甾醛作為原料,合成了4個(gè)具有不同結(jié)構(gòu)特征的N,N-二甲基甾體縮氨硫腙類化合物??鼓[瘤活性研究結(jié)果表明:所有化合物對(duì)所測(cè)試的腫瘤細(xì)胞都表現(xiàn)出明顯的生長增殖抑制活性。其中,具有17-縮氨硫腙結(jié)構(gòu)的化合物對(duì)人腎上皮正常細(xì)胞HEK-293T沒有表現(xiàn)出明顯的抑制活性。以上結(jié)果為新的甾體抗腫瘤藥物的設(shè)計(jì)合成提供參考。

Abstract:Four novel N,N-dimethylthiosemicarbazone compounds(1~4)were synthesized by condensation reaction of dehydroepiandrosterone,pregnenolone,estrone,3,5-dihydroxy-6-formyl-B-norcholestane with N,N-dimethylthiosemicarbazide,respectively,using EtOH(or MeOH)as solvent and HAc as catalyst.The structures were characterized by1H NMR,13C NMR,IR and HR-EI-MS.The anti-tumor activities of 1~4 against human hepatoma cells(HepG),nasopharyngeal carcinoma cells(CNE-2)and normal kidney epithelial cells(HEK293T)were investigated by MTT method.The results showed that 1~3 had better antiproliferative activities against HepG and CNE-2.
