昆蟲的泌蠟結(jié)構(gòu) (wax secreting structure) 是由昆蟲體壁外層表皮細(xì)胞特化形成的,包括泌蠟孔和泌蠟板2種形式,它們能將蠟腺細(xì)胞分泌的蠟質(zhì)排至體外,防止體內(nèi)水分過度蒸發(fā),保持體溫;蠟質(zhì)所形成的保護(hù)層還可以保護(hù)蟲體免受體外水汽侵入和真菌寄生[1-3]。泌蠟結(jié)構(gòu)主要存在于半翅目、膜翅目、鱗翅目、脈翅目昆蟲中[4-8]。蠟蟬總科 (Fulgoroidea)是半翅目昆蟲的重要類群,其成、若蟲都能分泌蠟質(zhì)[9]。目前已有學(xué)者對該類群的菱蠟蟬科(Cixidae)、脈蠟蟬科(Meenoplidae)、閹蠟蟬科(Kinnaridae)和瓢蠟蟬科(Issidae)等成、若蟲蠟絲、蠟腺或泌蠟結(jié)構(gòu)進(jìn)行了研究,并討論了其分類學(xué)價(jià)值[10-14],還有部分學(xué)者認(rèn)為不同蠟蟬類群成、若蟲的泌蠟結(jié)構(gòu)分布及形態(tài)特征可為不同類群的親緣關(guān)系探討提供重要依據(jù)[14-17]。
廣蠟蟬科(Ricaniidae)昆蟲成、若蟲均可分泌蠟質(zhì),其若蟲腹部6~9腹節(jié)可分泌出大量蠟絲,部分類群成蟲腹部末端分布有大量的蠟絲團(tuán)[18-19]。近年來,有學(xué)者對該科部分種類成、若蟲體表的蠟粉或蠟絲形態(tài)進(jìn)行了觀察描述,包括西藏疏廣蠟蟬(EuricaniaxizangensisChou&Lu[18])、圓紋寬廣蠟蟬[Pochaziaguttifera(Walker)[20]]、柿寬廣蠟蟬[Pochaziasublimata(Jacobi)[21]]和山東寬廣蠟蟬[Pochaziashantungensis(Chou & Lu)[22]]、八點(diǎn)廣翅蠟蟬[Ricaniaspeculum(Walker)[19,21,23]],但目前對該科昆蟲的成蟲泌蠟結(jié)構(gòu)尚無進(jìn)行深入探討。由此,本研究選擇該科中重要的植食性害蟲山東寬廣蠟蟬[Pochaziashantungensis(Chou & Lu,1977)]為研究對象,利用掃描電子顯微鏡 (SEM) 和顯微數(shù)碼成像相機(jī) (CCD) 對該種成蟲體表蠟粉、蠟絲以及泌蠟結(jié)構(gòu)進(jìn)行詳細(xì)觀察,旨在明確其泌蠟結(jié)構(gòu)及蠟絲形態(tài)特征,并探討廣蠟蟬科昆蟲的典型泌蠟結(jié)構(gòu),為該類群分類及系統(tǒng)發(fā)育研究提供更多的形態(tài)學(xué)資料。
試驗(yàn)材料均為成蟲,包括用體積分?jǐn)?shù)70%酒精浸泡的標(biāo)本和新鮮標(biāo)本,均由李健女士于2016年7月采自陜西楊凌西北農(nóng)林科技大學(xué)校園內(nèi)。
1.2.1 對成蟲標(biāo)本拍照 用展翅板將采集標(biāo)本進(jìn)行展翅,再在QImaging Retica 2000R顯微數(shù)碼成像相機(jī) (CCD) 對前、后翅和雌、雄蟲腹部進(jìn)行拍照。
1.2.2 電鏡觀察標(biāo)本處理 為了更好地觀察山東寬廣蠟蟬的泌蠟結(jié)構(gòu),本研究對采集的新鮮標(biāo)本和體積分?jǐn)?shù)70%酒精浸泡標(biāo)本分別采用除蠟和非除蠟2種方法進(jìn)行處理。
酒精浸泡標(biāo)本處理:方法1 (除蠟處理):將標(biāo)本放入氯仿中浸泡10 min除蠟,用體積分?jǐn)?shù)70%酒精沖洗2遍,再用體積分?jǐn)?shù)梯度酒精 (80%、90%、100% 各15 min) 脫水處理,取下所需觀察部位后置于自然條件下干燥;方法2 (非除蠟處理):將浸泡標(biāo)本直接放入體積分?jǐn)?shù)梯度酒精 (同上) 中進(jìn)行脫水處理,取下所需觀察部位后置于自然條件下干燥。
鮮標(biāo)本處理:方法1 (除蠟處理):將新鮮標(biāo)本放入氯仿中浸泡10 min,再放入體積分?jǐn)?shù)梯度酒精 (30%、40%、50%、60%、70%、80%、90%、100% 各15 min) 脫水處理,在自然條件下干燥;方法2 (非除蠟處理):將新鮮標(biāo)本進(jìn)行針插并展翅,再置于自然條件下干燥。
將干燥后的樣品用雙面導(dǎo)電膠粘臺(tái),用Hitachi MSP-IS (Japan,Tokyo) 進(jìn)行噴金處理,最后在Hitachi S-3400N掃描電子顯微鏡 (Japan,Tokyo) 下用15 kV進(jìn)行觀察并拍照。
在未除蠟情況下,山東寬廣蠟蟬前翅 (forewing) 正面有棕色蠟粉 (圖1-A、B),反面有淺黃色蠟粉 (圖2-A、B);后翅 (hind wing) 翅脈 (vein) 及翅脈兩側(cè)均有灰白色蠟粉 (圖3-A、B,圖4-A、B)。
2.1.1 前翅翅面泌蠟結(jié)構(gòu) 除蠟后的山東寬廣蠟蟬前翅正、反面均分布有大量氣孔狀蠟孔 (stomata-like wax pore),以及不規(guī)則突起 (irregularity bump) (圖1,圖2),蠟粉從氣孔狀蠟孔中間的泌蠟孔 (wax-secreting pore) 分泌出來 (圖1-E,圖2-E)。
前翅正面的氣孔狀蠟孔分布于翅脈 (圖1-C) 和翅面 (圖1-D)。該蠟孔由外緣 (raised outer edge),開口 (opening) 和泌蠟孔組成,外緣近橢圓形,開口呈梭形,泌蠟孔近圓形 (圖1-F,表1)。
前翅反面氣孔狀蠟孔僅分布在翅脈及翅脈兩側(cè) (圖2-C),翅前緣區(qū) (costal field) 泌蠟孔的密度大于其他區(qū)域 (圖2-C,表1)。

2.1.2 后翅翅面泌蠟結(jié)構(gòu) 除蠟后,發(fā)現(xiàn)山東寬廣蠟蟬后翅的氣孔狀蠟孔均僅分布于翅脈和翅脈兩側(cè) (圖3-C,圖4-C),翅其余部位無蠟孔,僅具不規(guī)則突起 (圖3-D,圖4-D),后翅正、反面蠟孔的形態(tài)、直徑差異較小(圖3-D、E,圖4-D、E)。后翅正面、反面氣孔狀蠟孔各部分量度見表1。
山東寬廣蠟蟬雌雄成蟲肩板 (tegula) 近四邊形 (圖5-A),正面分布有大量蠟孔(圖5-B),成蟲腹部體壁外層均分布有大量氣孔狀蠟孔 (圖5-E、F)。蠟孔形態(tài)及組成與翅面分布的蠟孔相同(圖5-C、D),包括外緣、開口以及泌蠟孔 (量度見表1)。
山東寬廣蠟蟬雌雄成蟲生殖節(jié) (genital segments) 形態(tài)差別明顯 (圖6-A、B、C、D)。雌成蟲腹部末端還有大量乳白色棉花團(tuán)狀蠟絲團(tuán) (雄成蟲腹部末端無)。對未除蠟干標(biāo)本進(jìn)行觀察,發(fā)現(xiàn)乳白色蠟絲由第9腹節(jié)背板(the ninth abdominal tergite) 和肛板(anal tube) 內(nèi)側(cè)的泌蠟板分泌形成 (圖6-E、F、G)。
2.3.1 第9腹節(jié)泌蠟結(jié)構(gòu) 第9腹節(jié)背板上分布著由十?dāng)?shù)根蠟絲 (直徑0.13 μm) 組成的蠟絲束 (直徑2.48~3.63 μm),在蠟絲束表面覆有大量蠟粉 (圖6-E、F),蠟絲束由位于背板上泌蠟板的泌蠟孔分泌,篩孔間隔 (sieve-pore partition wall) 表面覆有大量蠟粉 (圖6-E);第9腹節(jié)背板上的泌蠟板整體為長板狀篩板型泌蠟板 (sieve-plate wax secreting plate),泌蠟板中隨機(jī)分布有大量感覺毛 (sensory hair) (圖7-A)。泌蠟孔、近圓形底板 (orbicular base plate) 和不規(guī)則外緣 (irregularity edge) 組成一個(gè)標(biāo)準(zhǔn)泌蠟單元 (wax-secreting unit) (圖7-B、C、D)。

根據(jù)篩板間隔的寬度,可將泌蠟板分成3類 (表2):即闊型泌蠟板 (broad type of sieve-plate wax secreting plate) (圖7-B)、中型泌蠟板(medium type of sieve-plate wax secreting plate) (圖7-C) 和窄型泌蠟板(narrow type of sieve-plate wax secreting plate)(圖7-D),這3種泌蠟板的不規(guī)則外緣和篩孔間隔均分布大量的不規(guī)則凹陷 (irregularity hollow),闊型泌蠟板泌蠟單元外緣上方的凹陷淺于另外2種 (圖7-B、C、D)。以上3類泌蠟板泌蠟單元密度依次為1.4、1.89和2.3 U/100 μm2。
2.3.2 肛板內(nèi)側(cè)泌蠟結(jié)構(gòu) 山東寬廣蠟蟬雌蟲肛板內(nèi)側(cè)分布有大量乳白色蠟絲束,蠟絲束和泌蠟板表面覆有大量蠟粉。蠟絲束直徑2.73~3.67 μm,單根蠟絲直徑0.12 μm (圖6-G、H)。對經(jīng)過除蠟處理的雌蟲肛板觀察 (圖7),發(fā)現(xiàn)肛板內(nèi)側(cè)均為篩板狀泌蠟板,其內(nèi)側(cè)隨機(jī)分布數(shù)十根感覺毛 (圖7-E),1個(gè)標(biāo)準(zhǔn)泌蠟單元同樣由泌蠟孔、近圓形底板和不規(guī)則外緣組成 (圖7-F、G、H)。
依據(jù)篩孔間隔寬度,可將肛板內(nèi)側(cè)泌蠟板也分為3個(gè)類型 (表2),即闊型泌蠟板、中型泌蠟板和窄型泌蠟板。闊型泌蠟板出現(xiàn)在腹部未泌蠟的雌成蟲 (圖7-F),中型和窄型泌蠟板出現(xiàn)在腹部布滿蠟絲的雌成蟲 (圖7-F、G、H);闊型泌蠟板泌蠟單元外緣和篩孔間隔還布滿大量不規(guī)則深色斑點(diǎn) (圖7-F)。肛板內(nèi)側(cè)3種泌蠟板泌蠟單元密度依次為1.45、2.05、2.00 U/100 μm2。雄成蟲腹部第9節(jié)背板光滑,無泌蠟板 (圖7-I),肛板內(nèi)側(cè)無泌蠟板,僅分布大量鈍突(obtuse apophysis) (圖7-J、K)。

山東寬廣蠟蟬寄主范圍廣,包括大戟科(Euphorbiaceae)、???Moraceae)等分泌蜜露或樹膠的植物[24-27]。本研究發(fā)現(xiàn)該蟲成蟲前后翅、肩板以及腹部體壁外的氣孔狀蠟孔分泌蠟粉,這可能是該蟲在長期進(jìn)化過程中產(chǎn)生的一種適應(yīng)性進(jìn)化。分泌的蠟粉可能有助于避免寄主所分泌的蜜露或樹膠粘附蟲體從而影響其正常取食、飛行或交配等。此外,本研究還發(fā)現(xiàn)蟲體體表蠟粉分泌量變化較大,部分成蟲翅面及體軀上甚至沒有蠟粉 (據(jù)表1數(shù)據(jù),相同部位氣孔狀蠟孔開口的橫軸變化較大),可能是因?yàn)樯綎|寬廣蠟蟬成蟲在羽化初期氣孔狀蠟孔未發(fā)育完全,蠟孔開口較小,成蟲發(fā)育后期蠟孔發(fā)育完全,蠟孔開口變大,蠟粉分泌量隨之增多。
山東寬廣蠟蟬雌成蟲的第9腹節(jié)背板及肛板內(nèi)側(cè)的篩板狀泌蠟板均可分為闊型泌蠟板、中型泌蠟板和窄型泌蠟板,闊型泌蠟板出現(xiàn)在未泌蠟的雌成蟲腹部,而中型和窄型泌蠟板出現(xiàn)在已經(jīng)分泌蠟絲的雌成蟲腹部。結(jié)合圖7和表2數(shù)據(jù),本研究推測山東寬廣蠟蟬的泌蠟板發(fā)育可能經(jīng)過3個(gè)階段,闊型泌蠟板為泌蠟板的最初階段;雌成蟲交配后,泌蠟板的篩孔間隔 (sieve-pore partition wall) 上具有不規(guī)則凹痕的部分逐漸向泌蠟單元近圓形底板凹陷,形成中型泌蠟板;在產(chǎn)卵前期凹陷劇烈,直至篩孔間隔寬度至最小,不規(guī)則外緣發(fā)育至最大。此外,本研究還發(fā)現(xiàn):隨著山東寬廣蠟蟬腹部末端泌蠟板逐漸發(fā)育完全,蠟絲團(tuán)也因泌蠟單元密度的增大而變致密,這可能對蟲卵起到更好的保護(hù)作用。

蠟蟬總科昆蟲不同類群的成、若蟲體壁上的蠟孔或泌蠟板形態(tài)差異明顯,如蛾蠟蟬科(Flatidae)成蟲前翅爪區(qū)具獨(dú)特的念珠狀泌蠟板(moniliform glandular pore-plate),且該科內(nèi)不同屬的成蟲前翅念珠狀泌蠟板形態(tài)也有差異,可作為分類依據(jù)之一[28-29];閹蠟蟬科Kinnara屬與Adolenda屬雌成蟲腹部末端6~8節(jié)泌蠟板形態(tài)差異明顯,前者腹部泌蠟板被長脊劃分成不同單元格,形態(tài)上與科內(nèi)其他屬差異較大,是該屬特有的泌蠟結(jié)構(gòu)形態(tài),而后者腹部泌蠟板為多孔狀[13,30]。本研究通過觀察對比,發(fā)現(xiàn)山東寬廣蠟蟬蟲體氣孔狀蠟孔與與林江[21]所描述的柿寬廣蠟蟬(Pochaziasublimata)、八點(diǎn)廣翅蠟蟬(Ricaniaspeculum)和透明疏廣蠟蟬(EuricaniaclaraKato)前翅翅面蠟孔形態(tài)一致。據(jù)此本研究推測氣孔狀蠟孔可能是廣蠟蟬科昆蟲體表的典型泌蠟孔。
廣蠟蟬科是蠟蟬總科中比較獨(dú)特的一個(gè)科,成蟲前翅翅脈、翅斑及雄性外生殖器特征等是該類群常用的分類特征[9,31]。已有研究還發(fā)現(xiàn)一些廣蠟蟬的成、若蟲泌蠟結(jié)構(gòu)也存在差異,如:柿寬廣蠟蟬、八點(diǎn)廣翅蠟蟬及透明疏廣蠟蟬前翅正面蠟孔形態(tài)相似,但三者成蟲前翅蠟孔大小有差異,其中透明疏廣蠟蟬前翅蠟孔直徑最小[19];八點(diǎn)廣翅蠟蟬及柿寬廣蠟蟬若蟲泌蠟板數(shù)量隨齡期增長而遞增,后者蠟板數(shù)量隨齡期變化呈一階數(shù)列遞增,且二者5齡若蟲泌蠟板顏色存在差異[19,21]。此外,本文還研究了八點(diǎn)廣翅蠟蟬及圓紋寬廣蠟蟬的雌蟲生殖節(jié)泌蠟板(圖8),觀察比較后發(fā)現(xiàn):這3種廣蠟蟬的泌蠟板皆屬于篩板狀泌蠟板,但其泌蠟單元的組份及直徑存在差異,其中八點(diǎn)廣翅蠟蟬生殖節(jié)泌蠟板篩板間隔極大,泌蠟單元無不規(guī)則外緣;圓紋寬廣蠟蟬生殖節(jié)末端泌蠟板泌蠟孔突起,第9腹節(jié)背板泌蠟板泌蠟單元無不規(guī)則外緣,而肛板內(nèi)側(cè)泌蠟板泌蠟單元具不規(guī)則外緣;山東寬廣蠟蟬泌蠟板與圓紋寬廣蠟蟬泌蠟板形態(tài)更相似,可支持山東寬廣蠟蟬屬于寬廣蠟蟬屬。
蠟蟬總科若蟲泌蠟結(jié)構(gòu)的形態(tài)變化可以反映類群的進(jìn)化,分析不同類群若蟲泌蠟板的轉(zhuǎn)變模式有助于蠟蟬總科高級階元的系統(tǒng)關(guān)系研究[14]。目前已發(fā)現(xiàn)的特征包括5齡若蟲泌蠟板分布區(qū)域、形態(tài)特征和蠟絲形態(tài)等[11,17]。如通過比較利用雌成蟲腹部末端泌蠟板形態(tài)特征,發(fā)現(xiàn)脈蠟蟬科和閹蠟蟬科雌成蟲腹部生殖節(jié)上的泌蠟板具有同源性,可確定脈蠟蟬科和閹蠟蟬科為姊妹群[12,32]。廣蠟蟬科雌成蟲生殖節(jié)泌蠟板的形態(tài)與蛾蠟蟬科[28]、璐蠟蟬科[33]雌成蟲生殖節(jié)泌蠟板形態(tài)差異明顯,但三者的共同特征是泌蠟板分布于肛板內(nèi)側(cè)、泌蠟單元圓形、泌蠟單元中圓形泌蠟孔明顯,其腹部泌蠟板分布區(qū)域及形態(tài)與閹蠟蟬科[13]和菱蠟蟬科[34]不同,通過泌蠟板形態(tài)比較,推測廣蠟蟬科與蛾蠟蟬科、璐蠟蟬科更加近緣。結(jié)合前人研究,筆者認(rèn)為蠟蟬總科昆蟲的泌蠟結(jié)構(gòu)形態(tài)可以為該類群的分類及高級階元?jiǎng)澐痔峁┲匾螒B(tài)學(xué)資料。
