石家莊地區(qū)漢族人群18個STR基因座遺傳多態(tài)性調(diào)查

目的 調(diào)查石家莊漢族人群18個STR基因座的遺傳多態(tài)性。方法 應(yīng)用DNA TyperTM15 Plus熒光標(biāo)記復(fù)合擴(kuò)增系統(tǒng)檢測323名石家莊地區(qū)漢族無關(guān)個體18個STR基因座多態(tài)性,計算群體遺傳學(xué)參數(shù)。結(jié)果 18個STR基因型分布均符合Hardy-Weinberg平衡(P>0.05),共檢出198個等位基因, 695個基因型,12個微變異等位基因。結(jié)論 18個STR基因座在石家莊漢族人群中有較高的遺傳多態(tài)性。

應(yīng)用DNA TyperTM15 Plus擴(kuò)增試劑盒(公安部物證鑒定中心)對石家莊地區(qū)323名漢族無關(guān)人群18個基因座(CSF1PO、D2S1338、D3S1358、D5S818、D6S1043、D7S820、D8S1179、D12S391、D13S317、D16S539、D18S51、D19S433、D21S11、 FGA、PentaE、TH01、TPOX、vWA)進(jìn)行調(diào)查,用Modified-Powerstates分析軟件進(jìn)行統(tǒng)計學(xué)計算[1],應(yīng)用Hardy-Weinberg平衡軟件對各個基因座數(shù)據(jù)進(jìn)行χ2檢驗[2],獲得了該地區(qū)漢族群體遺傳學(xué)數(shù)據(jù)(見表1、表2)。

18個STR基因座,共檢測到198個等位基因,其頻率在0.002~0.545之間,共695個基因型。在8個基因座上檢出12個微變異等位基因(OL),其頻率均小于0.005(見表3),對于法醫(yī)的個體識別及親子鑒定具有一定的意義。各基因座的H值分布在0.624~0.932之間,DP值0.811~0.984,PE值0.321~0.861,PIC值0.580~0.910,累計匹配率為5.736×10-22,累計個體識別率達(dá)到0.999 999 999 999 999 999 999 426,三聯(lián)體的累計非父排除概率0.999 999 973 339。

Genetic Polymorphism of 18 STR Loci of Han Population in Shijiazhuang

Objective To investigate genetic polymorphism of 18 STR loci on human chromosomes of Han population in Shijiazhuang. Methods Allelic frequencies for 18 STR loci and genetic parameters for forensic purpose were calculated. Results No deviations of allelic frequency from Hardy-Weinberg equilibrium expectations were observed (P>0.05). 198 alleles and 695 genotypes were gained. Allele frequencies varied between 0.002 and 0.545; heterozygosity (H) between 0.624 and 0.932; matching probability (Pm) between 0.016 and 0.189; power of discrimination (DP) between 0.811 and 0.984; polymorphism information content (PIC) between 0.580 and 0.910; power exclusion (PE) between 0.321 and 0.861. 12 off-ladder alleles of 8 STR loci were found. Conclusions 18 STR loci of Han population in Shijiazhuang are with relatively high polymorphism, and suitable for forensic individual identifi cation, paternity testing, and relative research.
